Présentation

Les douleurs chroniques affectent environ 20% des adultes et jusqu’à 50% de la population âgée. Cependant, les grandes familles d’antalgiques disponibles sont souvent d’une efficacité limitée et/ou possèdent une mauvaise tolérance, des effets secondaires importants ou des risques d’abus. Malgré cela, la thérapeutique médicamenteuse de la douleur n’a connu que peu d’innovations au cours des dernières années à cause d’une connaissance encore très incomplète de la physiopathologie associée. La découverte de nouvelles cibles moléculaires représente ainsi une réelle opportunité pour développer de nouvelles stratégies contre la douleur.

Parmi les senseurs moléculaires de la douleur récemment identifiés figurent les canaux ioniques ASIC (Acid Sensing Ion Channel), qui forment une classe de canaux cationiques dépolarisants excitateurs activés par les protons extracellulaires, et les canaux K2P, qui forment une classe de canaux potassiques hyperpolarisants inhibiteurs.

Le projet développé au sein de l’équipe a pour but d’étudier les propriétés et le rôle de ces canaux dans la douleur en combinant des approches de biologie moléculaire et cellulaire, d’électrophysiologie, de pharmacologie incluant la découverte et l’utilisation de peptides issus de venins et ciblant spécifiquement ces canaux, et des études de comportement chez l’animal.
