Some amphipathic helices exhibit unique binding properties due to their specific amino acid composition. For example, the ALPS motif, which detects membrane curvature, is characterized by the abundance of small, uncharged residues on its polar face. We are investigating how the amino acid composition of other amphipathic helices determines their binding properties. A recent example is perilipin-4, a giant helix that binds directly to triglyceride droplets but not to lipid-phospholipid bilayers. in vitro.
