Within a clonal population, a single drug acts on a continuum of pre-existing cellular sensitivity states. The dose repositions a decision surface that partitions this space into sensitive and tolerant zones. The observed efficacy thus results from the interaction between the drug and the distribution of molecular states present at the time of treatment.
